Een fietskoerier uit New York (Joseph Gordon-Levitt) krijgt een envelop van een jonge vrouw op een universiteit in een buitenwijk van Manhattan en hem wordt verteld dat hij 90 minuten heeft om de envelop te bezorgen op een adres in Chinatown. Complicaties ontstaan wanneer een undercover politieagent verschijnt en de envelop onder speciale omstandigheden opeist. De waarheid, verborgen motieven en de inzet op leven en dood worden onthuld aan de hand van een reeks flashbacks. Dit terwijl de agent en de boodschapper in een hevige achtervolging door heel Manhattan verwikkeld raken, racend tegen de klok nadat de boodschapper de kostbare aard ontdekt van de geringe inhoud van de envelop.
