Omdat de bloedmooie Maria uit hun leven verdwijnt door naar Amsterdam te vertrekken, besluiten de twee vrienden Harold Lee en Kumar Patel haar achterna te gaan. Maar onderweg naar Amsterdam probeert het tweetal een grote waterpijp mee te smokkelen. Het duurt niet lang voordat de passagiers in het vliegtuig de twee aanzien voor terroristen. Vanaf dat moment komen Harold en Kumar in de meest bizarre situaties terecht.
