In San Francisco heeft Jill Valentine te maken met een zombie-uitbraak en een nieuw T-Virus, Leon Kennedy volgt een ontvoerde DARPA-wetenschapper op het spoor en Claire Redfield onderzoekt een monsterlijke vis die walvissen doodt in de baai. Samen met Chris Redfield en Rebecca Chambers ontdekken ze het spoor van aanwijzingen uit hun afzonderlijke zaken die allemaal samenkomen op dezelfde locatie, Alcatraz Island, waar een nieuw kwaad zijn intrek heeft genomen en op hun komst wacht.
