Professor Sherman Klump probeert een wit voetje te halen bij zijn nieuwe collega Denise. Na een mislukt DNA-experiment worden Sherman en zijn alter ego Buddy Love van elkaar gescheiden. Een catastrofe voor de brave Sherman die elementen uit Denises onderzoek nodig heeft om voor eens en altijd af te rekenen met Buddy Love.
