Acchi Kocchi vertelt over de alledaagse waanzin van de vijf klasgenoten: Tsumiki, op het eerste gezicht een klein en gereserveerd meisje, maar dat haar emoties soms niet helemaal kan dragen of bedwingen zoals ze zou willen; Io is een brildragende en stoere allrounder die de neuzen van alle meisjes doet bloeden; Hime, het lieve, beleefde en goedhartige meisje van de groep; Mayoi, de grappige in de laboratoriumjas, wiens hoofd helaas is volgepropt met te veel waanzin en gekte en Sakaki, de grappenmaker, die qua aard het beste bij Mayoi past.
