FBI-agent Malcolm Turner is op jacht naar een ontsnapte bankrover die vermoedelijk naar het huis van Big Momma zal gaan, de grootmoeder van diens knappe ex-vriendin Sherry. Wanneer Big Momma plotseling op reis gaat, dreigt het plan te mislukken en is het aan Malcolm om de rol van de spraakmakende oma over te nemen. Malcolm heeft alles onder controle, behalve wanneer hij verliefd wordt op Sherry.
